Uncertain significance for Spastic ataxia 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006612.6(KIF1C):c.3239del (p.Pro1080fs), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with KIF1C-related conditions. The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the ExAC database. This sequence change creates a premature translational stop signal (p.Pro1080Hisfs*8) in the KIF1C gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 24 amino acid(s) of the KIF1C protein. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
