Uncertain significance for Congenital glucose-galactose malabsorption — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000343.4(SLC5A1):c.1403C>A (p.Ala468Glu), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals with SLC5A1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C65"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant is present in population databases (rs200406921, ExAC 0.001%). This sequence change replaces alanine with glutamic acid at codon 468 of the SLC5A1 protein (p.Ala468Glu). The alanine residue is highly conserved and there is a moderate physicochemical difference between alanine and glutamic acid.
