Uncertain significance for 3MC syndrome 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_139125.4(MASP1):c.491A>G (p.Tyr164Cys), citing Invitae Variant Classification Sherloc (09022015): This variant is present in population databases (rs535846193, ExAC 0.1%). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant has not been reported in the literature in individuals with MASP1-related conditions. This sequence change replaces tyrosine with cysteine at codon 164 of the MASP1 protein (p.Tyr164Cys). The tyrosine residue is moderately conserved and there is a large physicochemical difference between tyrosine and cysteine.
