Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_206933.4(USH2A):c.1252A>T (p.Asn418Tyr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces asparagine with tyrosine at codon 418 of the USH2A protein (p.Asn418Tyr). The asparagine residue is highly conserved and there is a large physicochemical difference between asparagine and tyrosine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This variant has not been reported in the literature in individuals affected with USH2A-related conditions. This variant is not present in population databases (ExAC no frequency).
