Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_007186.6(CEP250):c.4918G>A (p.Glu1640Lys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid with lysine at codon 1640 of the CEP250 protein (p.Glu1640Lys). The glutamic acid residue is weakly conserved and there is a small physicochemical difference between glutamic acid and lysine. This variant is present in population databases (rs755541063, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with CEP250-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Not Available"; PolyPhen-2: "Benign"; Align-GVGD: "Not Available". The lysine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function.
