Pathogenic for Essential thrombocythemia; Congenital amegakaryocytic thrombocytopenia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005373.3(MPL):c.605dup (p.Ala203fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the MPL gene (transcript NM_005373.3) at coding-DNA position 605, duplicating one base; at the protein level this means shifts the reading frame starting at alanine residue 203, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with MPL-related conditions. This variant is present in population databases (no rsID available, gnomAD 0.006%). This sequence change creates a premature translational stop signal (p.Ala203Serfs*27) in the MPL gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in MPL are known to be pathogenic (PMID: 8073287, 11133753).
