This variant lies in the SDHC gene (transcript NM_003001.5) at coding-DNA position 475, where A is replaced by C; at the protein level this means replaces threonine at residue 159 with proline — a missense variant. Submitter rationale: This sequence change replaces threonine with proline at codon 159 of the SDHC protein (p.Thr159Pro). The threonine residue is weakly conserved and there is a small physicochemical difference between threonine and proline. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
