Uncertain significance for Hyperekplexia 3 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004211.5(SLC6A5):c.469A>G (p.Ser157Gly), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine with glycine at codon 157 of the SLC6A5 protein (p.Ser157Gly). The serine residue is moderately conserved and there is a small physicochemical difference between serine and glycine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with SLC6A5-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt SLC6A5 protein function.
