Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_172351.3(CD46):c.424G>C (p.Glu142Gln), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid, which is acidic and polar, with glutamine, which is neutral and polar, at codon 142 of the CD46 protein (p.Glu142Gln). This variant is present in population databases (rs371106885, gnomAD 0.02%). This missense change has been observed in individual(s) with atypical hemolytic uremic syndrome (PMID: 21906045, 25733390, 28461395). ClinVar contains an entry for this variant (Variation ID: 1381088). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt CD46 protein function with a negative predictive value of 80%.
