This variant has been observed in at least one proband affected with atypical hemolytic-uremic syndrome (PMID:25862562;25733390;21906045;26826462;28461395). Functional studies have been reported; however, the significance of the findings remain unclear (PMID:25733390). It is absent or not present at a significant frequency in gnomAD. In conclusion, we classify CD46 p.Glu142Gln (c.424G>C) as a variant of uncertain significance.
