NM_000321.3(RB1):c.1802C>G (p.Thr601Ser) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.T601S variant (also known as c.1802C>G), located in coding exon 18 of the RB1 gene, results from a C to G substitution at nucleotide position 1802. The threonine at codon 601 is replaced by serine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
