Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000275.3(OCA2):c.470A>T (p.Asp157Val), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (ExAC no frequency). This sequence change replaces aspartic acid with valine at codon 157 of the OCA2 protein (p.Asp157Val). The aspartic acid residue is weakly conserved and there is a large physicochemical difference between aspartic acid and valine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt OCA2 protein function. This variant has not been reported in the literature in individuals affected with OCA2-related conditions.
