This variant lies in the SDHB gene (transcript NM_003000.3) at coding-DNA position 183, where T is replaced by A; at the protein level this means converts the codon for tyrosine at residue 61 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This variant is not present in population databases (gnomAD no frequency). For these reasons, this variant has been classified as Pathogenic. This premature translational stop signal has been observed in individuals with SDHB-related conditions (PMID: 20418362, 31492822; Invitae). This sequence change creates a premature translational stop signal (p.Tyr61*) in the SDHB gene. It is expected to result in an absent or disrupted protein product.
