Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007194.4(CHEK2):c.421A>G (p.Lys141Glu), citing Ambry Variant Classification Scheme 2023: The p.K141E variant (also known as c.421A>G), located in coding exon 2 of the CHEK2 gene, results from an A to G substitution at nucleotide position 421. The lysine at codon 141 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
