NM_020376.4(PNPLA2):c.1427C>A (p.Ser476Tyr) was classified as Uncertain significance for Neutral lipid storage myopathy by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine with tyrosine at codon 476 of the PNPLA2 protein (p.Ser476Tyr). The serine residue is weakly conserved and there is a large physicochemical difference between serine and tyrosine. The frequency data for this variant in the population databases is considered unreliable, as metrics indicate insufficient coverage at this position in the ExAC database. This variant has not been reported in the literature in individuals affected with PNPLA2-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated.
