Uncertain significance for Autosomal recessive polycystic kidney disease — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_138694.4(PKHD1):c.7414A>G (p.Thr2472Ala), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces threonine with alanine at codon 2472 of the PKHD1 protein (p.Thr2472Ala). The threonine residue is highly conserved and there is a small physicochemical difference between threonine and alanine. This variant is present in population databases (rs137972951, ExAC 0.001%). This variant has not been reported in the literature in individuals affected with PKHD1-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt PKHD1 protein function.
