Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt COL4A2 protein function. This variant has not been reported in the literature in individuals with COL4A2-related conditions. This variant is present in population databases (rs779464248, ExAC 0.05%). This sequence change replaces leucine with phenylalanine at codon 1099 of the COL4A2 protein (p.Leu1099Phe). The leucine residue is highly conserved and there is a small physicochemical difference between leucine and phenylalanine.
