Uncertain significance for Lethal congenital glycogen storage disease of heart — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_016203.4(PRKAG2):c.1392T>A (p.Asp464Glu), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with PRKAG2-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces aspartic acid with glutamic acid at codon 464 of the PRKAG2 protein (p.Asp464Glu). The aspartic acid residue is highly conserved and there is a small physicochemical difference between aspartic acid and glutamic acid. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0").
