This variant lies in the PLEKHG5 gene (transcript NM_020631.6) at coding-DNA position 535, where C is replaced by T; at the protein level this means replaces proline at residue 179 with serine — a missense variant. Submitter rationale: This sequence change replaces proline with serine at codon 179 of the PLEKHG5 protein (p.Pro179Ser). The proline residue is weakly conserved and there is a moderate physicochemical difference between proline and serine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with PLEKHG5-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated.
