This variant lies in the GUCY2D gene (transcript NM_000180.4) at coding-DNA position 2842, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 948 with lysine — a missense variant. Submitter rationale: In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This missense change has been observed in individual(s) with Leber congenital amaurosis (Invitae). In at least one individual the data is consistent with the variant being in trans (on the opposite chromosome) from a pathogenic variant. This variant is not present in population databases (ExAC no frequency). This sequence change replaces glutamic acid with lysine at codon 948 of the GUCY2D protein (p.Glu948Lys). The glutamic acid residue is highly conserved and there is a small physicochemical difference between glutamic acid and lysine.
