This variant lies in the PIGO gene (transcript NM_032634.4) at coding-DNA position 1333, where C is replaced by T; at the protein level this means replaces arginine at residue 445 with cysteine — a missense variant. Submitter rationale: This sequence change replaces arginine with cysteine at codon 445 of the PIGO protein (p.Arg445Cys). The arginine residue is highly conserved and there is a large physicochemical difference between arginine and cysteine. This variant is present in population databases (rs779260324, ExAC 0.009%). This variant has not been reported in the literature in individuals affected with PIGO-related conditions.
