This variant lies in the COG5 gene (transcript NM_006348.5) at 12 bases upstream of the translation start (5' untranslated region), where G is replaced by A. Submitter rationale: This sequence change replaces alanine with threonine at codon 28 of the COG5 protein (p.Ala28Thr). The alanine residue is weakly conserved and there is a small physicochemical difference between alanine and threonine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with COG5-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated.
