NM_001277115.2(DNAH11):c.11847A>C (p.Arg3949Ser) was classified as Uncertain significance for Primary ciliary dyskinesia by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R3949S variant (also known as c.11847A>C), located in coding exon 73 of the DNAH11 gene, results from an A to C substitution at nucleotide position 11847. The arginine at codon 3949 is replaced by serine, an amino acid with dissimilar properties. This nucleotide position is not well conserved in available vertebrate species. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
