Uncertain significance for Leber congenital amaurosis 8; Retinitis pigmentosa 12 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_201253.3(CRB1):c.1958T>G (p.Ile653Ser), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (ExAC no frequency). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt CRB1 protein function. This missense change has been observed in individual(s) with retinitis pigmentosa (Invitae). This sequence change replaces isoleucine with serine at codon 653 of the CRB1 protein (p.Ile653Ser). The isoleucine residue is highly conserved and there is a large physicochemical difference between isoleucine and serine.
