Uncertain significance for Leber congenital amaurosis 13 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_152443.3(RDH12):c.782G>C (p.Gly261Ala), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with RDH12-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant is not present in population databases (ExAC no frequency). This sequence change replaces glycine with alanine at codon 261 of the RDH12 protein (p.Gly261Ala). The glycine residue is highly conserved and there is a small physicochemical difference between glycine and alanine.
