Uncertain significance for Dyskeratosis congenita — the classification assigned by Ambry Genetics to NM_198253.3(TERT):c.1349A>G (p.Gln450Arg), citing Ambry Variant Classification Scheme 2023: The p.Q450R variant (also known as c.1349A>G), located in coding exon 2 of the TERT gene, results from an A to G substitution at nucleotide position 1349. The glutamine at codon 450 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
