Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000179.3(MSH6):c.3387T>G (p.Cys1129Trp), citing Ambry Variant Classification Scheme 2023: The p.C1129W variant (also known as c.3387T>G), located in coding exon 5 of the MSH6 gene, results from a T to G substitution at nucleotide position 3387. The cysteine at codon 1129 is replaced by tryptophan, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
