This variant lies in the TTN gene (transcript NM_001267550.2) at coding-DNA position 105000, where A is replaced by T; at the protein level this means replaces glutamic acid at residue 35000 with aspartic acid — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function are unavailable for the TTN gene. This sequence change replaces glutamic acid with aspartic acid at codon 35000 of the TTN protein (p.Glu35000Asp). There is a small physicochemical difference between glutamic acid and aspartic acid. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with TTN-related disease.
