Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_030928.4(CDT1):c.1339C>T (p.Arg447Trp), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine with tryptophan at codon 447 of the CDT1 protein (p.Arg447Trp). The arginine residue is highly conserved and there is a moderate physicochemical difference between arginine and tryptophan. This variant is present in population databases (rs755342796, ExAC 0.006%). This variant has not been reported in the literature in individuals affected with CDT1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0").
