Uncertain significance for Charcot-Marie-Tooth disease axonal type 2P — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001005373.4(LRSAM1):c.2096G>T (p.Cys699Phe), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces cysteine with phenylalanine at codon 699 of the LRSAM1 protein (p.Cys699Phe). The cysteine residue is highly conserved and there is a large physicochemical difference between cysteine and phenylalanine. This variant is not present in population databases (ExAC no frequency). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This variant has not been reported in the literature in individuals affected with LRSAM1-related conditions.
