This variant lies in the STXBP2 gene (transcript NM_006949.4) at coding-DNA position 1079, where C is replaced by T; at the protein level this means replaces serine at residue 360 with leucine — a missense variant. Submitter rationale: This sequence change replaces serine with leucine at codon 360 of the STXBP2 protein (p.Ser360Leu). The serine residue is moderately conserved and there is a large physicochemical difference between serine and leucine. This variant is present in population databases (rs779452337, ExAC 0.006%). This variant has not been reported in the literature in individuals affected with STXBP2-related conditions.
