This variant lies in the HSPB1 gene (transcript NM_001540.5) at coding-DNA position 530, where A is replaced by T; at the protein level this means replaces asparagine at residue 177 with isoleucine — a missense variant. Submitter rationale: This sequence change replaces asparagine with isoleucine at codon 177 of the HSPB1 protein (p.Asn177Ile). The asparagine residue is weakly conserved and there is a large physicochemical difference between asparagine and isoleucine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with HSPB1-related conditions.
