Uncertain significance for Autosomal recessive spinocerebellar ataxia 12; Developmental and epileptic encephalopathy, 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_016373.4(WWOX):c.287A>T (p.Asp96Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces aspartic acid with valine at codon 96 of the WWOX protein (p.Asp96Val). The aspartic acid residue is highly conserved and there is a large physicochemical difference between aspartic acid and valine. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with WWOX-related conditions. ClinVar contains an entry for this variant (Variation ID: 1371374). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
