Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001077446.4(TSEN34):c.803C>T (p.Thr268Ile), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces threonine with isoleucine at codon 268 of the TSEN34 protein (p.Thr268Ile). The threonine residue is weakly conserved and there is a moderate physicochemical difference between threonine and isoleucine. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with TSEN34-related conditions. ClinVar contains an entry for this variant (Variation ID: 1371080). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
