This variant lies in the MYH7 gene (transcript NM_000257.4) at the canonical splice acceptor site of the intron immediately before coding-DNA position 531, where A is replaced by G; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This variant is not present in population databases (ExAC no frequency). This sequence change affects an acceptor splice site in intron 6 of the MYH7 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), however the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in MYH7 cause disease. This variant has not been reported in the literature in individuals affected with MYH7-related conditions.
