Uncertain significance for Joubert syndrome 25 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014704.4(CEP104):c.286G>C (p.Gly96Arg), citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This variant has not been reported in the literature in individuals affected with CEP104-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces glycine with arginine at codon 96 of the CEP104 protein (p.Gly96Arg). The glycine residue is highly conserved and there is a moderate physicochemical difference between glycine and arginine.
