Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.6133C>A (p.Pro2045Thr), citing Ambry Variant Classification Scheme 2023: The p.P2045T variant (also known as c.6133C>A), located in coding exon 44 of the POLE gene, results from a C to A substitution at nucleotide position 6133. The proline at codon 2045 is replaced by threonine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
