This variant lies in the COL4A1 gene (transcript NM_001845.6) at coding-DNA position 2705, where C is replaced by T; at the protein level this means replaces proline at residue 902 with leucine — a missense variant. Submitter rationale: Variant summary: COL4A1 c.2705C>T (p.Pro902Leu) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 8e-06 in 251384 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.2705C>T has been reported in the literature as a VUS in an individual with microcephaly (Ganapathy_2019) and in an individual affected by recurrent intracerebral hemorrhages, with an onset at 71 years of age (Liu_2022). These reports do not provide unequivocal conclusions about association of the variant with Porencephaly 1. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 31069529, 35711275). Three submitters have cited clinical-significance assessments for this variant to ClinVar after 2014. All submitters classified the variant as uncertain significance.
