Pathogenic for Microcephaly, normal intelligence and immunodeficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002485.5(NBN):c.1523dup (p.Ser509fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Ser509Ilefs*2) in the NBN gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in NBN are known to be pathogenic (PMID: 9590180, 16415040). This variant is not present in population databases (ExAC no frequency). This premature translational stop signal has been observed in individual(s) with prostate cancer (PMID: 31214711). For these reasons, this variant has been classified as Pathogenic.
