Pathogenic for Early-infantile DEE — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006030.4(CACNA2D2):c.2920_2921del (p.Leu974fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Leu974Valfs*103) in the CACNA2D2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in CACNA2D2 are known to be pathogenic (PMID: 24358150). This variant is not present in population databases (gnomAD no frequency). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. ClinVar contains an entry for this variant (Variation ID: 1369986). This variant has not been reported in the literature in individuals affected with CACNA2D2-related conditions. For these reasons, this variant has been classified as Pathogenic.
