NM_000334.4(SCN4A):c.2711C>A (p.Pro904Gln) was classified as Uncertain significance for Hyperkalemic periodic paralysis by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (ExAC no frequency). This sequence change replaces proline with glutamine at codon 904 of the SCN4A protein (p.Pro904Gln). The proline residue is moderately conserved and there is a moderate physicochemical difference between proline and glutamine. This variant has not been reported in the literature in individuals with SCN4A-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
