Pathogenic for Familial hypobetalipoproteinemia 1; Hypercholesterolemia, autosomal dominant, type B — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000384.3(APOB):c.2979T>A (p.Tyr993Ter), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals with APOB-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change creates a premature translational stop signal (p.Tyr993*) in the APOB gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in APOB are known to be pathogenic (PMID: 20032471).
