This variant lies in the CDH1 gene (transcript NM_004360.5) at coding-DNA position 1238, where A is replaced by G; at the protein level this means replaces tyrosine at residue 413 with cysteine — a missense variant. Submitter rationale: This sequence change replaces tyrosine with cysteine at codon 413 of the CDH1 protein (p.Tyr413Cys). The tyrosine residue is highly conserved and there is a large physicochemical difference between tyrosine and cysteine. This variant is not present in population databases (ExAC no frequency).
