This variant lies in the ADAR gene (transcript NM_001111.5) at coding-DNA position 7, where C is replaced by G; at the protein level this means replaces proline at residue 3 with alanine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). This variant has not been reported in the literature in individuals with ADAR-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces proline with alanine at codon 3 of the ADAR protein (p.Pro3Ala).
