Uncertain significance for Hereditary nonpolyposis colorectal neoplasms — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000179.3(MSH6):c.3173A>T (p.Asp1058Val), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the MSH6 gene (transcript NM_000179.3) at coding-DNA position 3173, where A is replaced by T; at the protein level this means replaces aspartic acid at residue 1058 with valine — a missense variant. Submitter rationale: In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This variant has not been reported in the literature in individuals affected with MSH6-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces aspartic acid with valine at codon 1058 of the MSH6 protein (p.Asp1058Val). The aspartic acid residue is highly conserved and there is a large physicochemical difference between aspartic acid and valine.
