NM_018127.7(ELAC2):c.631C>T (p.Arg211Ter) was classified as Uncertain significance for Combined oxidative phosphorylation defect type 17 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Arg211*) in the ELAC2 gene. However, it is currently unclear if variants that occur in this region of the gene cause disease. This variant is present in population databases (rs761881815, gnomAD 0.007%). This premature translational stop signal has been observed in individual(s) with hypertrophic cardiomyopathy (PMID: 23849775). It has also been observed to segregate with disease in related individuals. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
