Uncertain significance for Developmental and epileptic encephalopathy, 34 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_020708.5(SLC12A5):c.1775G>A (p.Arg592Gln), citing Invitae Variant Classification Sherloc (09022015): Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt SLC12A5 protein function. This sequence change replaces arginine with glutamine at codon 592 of the SLC12A5 protein (p.Arg592Gln). The arginine residue is moderately conserved and there is a small physicochemical difference between arginine and glutamine. This variant is present in population databases (rs368661669, ExAC 0.02%). This variant has not been reported in the literature in individuals with SLC12A5-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
