Uncertain significance for Mitochondrial DNA depletion syndrome, encephalomyopathic form with methylmalonic aciduria — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003850.3(SUCLA2):c.8C>T (p.Ala3Val), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with SUCLA2-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant is not present in population databases (ExAC no frequency). This sequence change replaces alanine with valine at codon 3 of the SUCLA2 protein (p.Ala3Val). The alanine residue is moderately conserved and there is a small physicochemical difference between alanine and valine.
