Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001349884.2(DRAM2):c.3G>A (p.Met1Ile), citing Invitae Variant Classification Sherloc (09022015): This sequence change affects the initiator methionine of the DRAM2 mRNA. The next in-frame methionine is located at codon 59. This variant is present in population databases (no rsID available, gnomAD 0.002%). Disruption of the initiator codon has been observed in individual(s) with clinical features of cone-rod dystrophy (PMID: 29555955, 35806404). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 1367200). This variant disrupts a region of the DRAM2 protein in which other variant(s) (p.Asp45Asn) have been observed in individuals with DRAM2-related conditions (Invitae). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic.
