Uncertain significance for Neuronopathy, distal hereditary motor, autosomal recessive 4; Charcot-Marie-Tooth disease recessive intermediate C — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_020631.6(PLEKHG5):c.2312C>A (p.Ser771Tyr), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (ExAC no frequency). This sequence change replaces serine with tyrosine at codon 771 of the PLEKHG5 protein (p.Ser771Tyr). The serine residue is moderately conserved and there is a large physicochemical difference between serine and tyrosine. This variant has not been reported in the literature in individuals affected with PLEKHG5-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
