Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_033004.4(NLRP1):c.164C>T (p.Ser55Leu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine, which is neutral and polar, with leucine, which is neutral and non-polar, at codon 55 of the NLRP1 protein (p.Ser55Leu). This variant is present in population databases (rs201750573, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with NLRP1-related conditions. ClinVar contains an entry for this variant (Variation ID: 1366760). An algorithm developed to predict the effect of missense changes on protein structure and function outputs the following: PolyPhen-2: "Possibly Damaging". The leucine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. Experimental studies have shown that this missense change does not substantially affect NLRP1 function (PMID: 27662089). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
